Firefly luciferase fusion

In this chapter we provide a detailed method for using Xenopus egg extract preparations that are optimized for examining protein turnover. We take advantage of firefly luciferase (Luciferase), a protein normally stable in Xenopus egg extract that, when fused to proteins of interest, provides a simple and rapid readout of protein turnover. We describe herein how these Luciferase fusion proteins can be used to perform high-throughput (HTS) biochemical screens in Xenopus egg extract to identify biologically active small-molecule compounds. [Pg.64]
