Experiment 64 Ultraviolet Spectra of Nucleotides

All nucleotides absorb electromagnetic radiation in the ultraviolet region, specifically between 200 and 

300 nm. Spectra of nucleotides obtained from a spectrophotometer maybe useful in identifying nucleotides. 

Prepare a 10 mM solution of K3P04 buffer. Use this solution as a blank and for diluting solutions of the standards. Prepare standard solutions of selected nucleotides and nucleoside triphosphates, 

10 mM in 10 mM K3P04 buffer. Obtain unknown nucleotide or nucleoside triphosphate solutions from your instructor. 

Zero the spectrophotometer at 300 nm, using the blank. Obtain molecular absorption spectra of all nucleotide and nucleoside solutions in the range of 200 to 300 nm.
