Disposition in the Body. Rapidly and almost completely absorbed after oral administration. It is metabolised mainly by hydroxy-lation. The major metabolite, 1-hydroxymethyltriazolam, has a hypnotic potency approximately equivalent to that of triazolam. Other metabolites include 4-hydroxytriazolam, 1-hydroxy-methyl-4-hydroxytriazolam, and three dichlorotriazolylbenzo-phenone derivatives. About 80% of a dose is excreted in the urine in 48 hours and about 7% is eliminated in the faeces in 72 hours.

Triazolam is absorbed well orally, exerts its sedative effects in 15 minutes, is distributed widely in the body, is bound to plasma proteins to the extent of 90%, is metabolized to 6-hydroxytriazolam, which is subsequently conjugated with glucuronic acid and is excreted in the urine. 

Nefazodone appears to inhibit the oxidative metabolism of alprazolam, midazolam, triazolam and zopiclone by the cytochrome P450 isoenzyme CYP3A4 so that they accumulate in the body. Lorazepam is unaffected because it is primarily excreted as a conjugate.
