It was synthesized from p-iodoanisole (65) by copper-catalyzed coupling with p-trifluoromethyliodobenzene (66) to give the expected statistical mixture from which unsymmetrical product 67 could be separated. Ether cleavage with HBr and HOAc gave 68 this was then alkylated with the aziridinium ion derived from N-(2-chloroethyl)pyrrolidine, using NaH as base, to complete the synthesis of boxidine (69). 

These eryt/ira-nitroaldols are attractive precursors to 1,2-amino alcohols (3). The conversion can be effected with retention of configuration by reduction to 2 (H, and neutral Raney nickel) followed by silyl ether cleavage with lithium aluminum hydride in ether. A one-step reduction of 1 to a 1,2-amino alcohol can be effected with lithium aluminum hydride, but with loss of configurational purity and some C—C bond cleavage. 

Standard peptide coupling chemistry is performed (Box 25). The acid component is transformed to an activated ester derivative and then treated with the amine [17-19]. This procedure is repeated twice. In the final step the ligands la-le-H4 are deprotected by aryl ether cleavage with BBr3. By use of this simple reaction sequence derivatives with glycine, alanine, phenylalanine, valine, leucine, and other amino acids as spacers were obtained [16].
