The left side summarizes the enzyme-dependent terms turnover number kcat and surface concentration renz- In case of enzyme-loaded films, renz should be replaced by the product of enzyme volume concentration in the film and the film thickness. The right side summarizes the experimental conditions diffusion coefficient D, concentration c of the mediator, and UME radius rT. The feedback mode always requires an as small as possible working distance d. The smaller the UME the more difficult it will be to detect the activity of the immobilized enzyme.

The half life of Superscript n is 30-60min at 37°C therefore, long incubations require extra enzyme to be added periodically. But it is advisable to keep the enzyme volume to less than 10% of the total reaction volume, as higher glycerol contents may interfere with reaction efficiency. 

Mix the DNA with 2 pL of the appropriate buffer (see Table 1) in a 500-pL microfuge tube. Add 1 pL of each enzyme (total enzyme volume cannot exceed 10% of reaction volume). Increase the volume to 20 pL with dH O.
