3 Molecular Dynamics of anti-DNP Antibody Binding Site [Pg.292]

A combined fluorescence-photochrome approach was used for investigating the molecular dynamics of anti-DNP antibody binding site and its cavity. A 4-(N-2,4-dinitrophenylamino)-4 -(N,N -dimethylamino)stilbene (StDNP) fluorescence DNP analogue was incorporated into the antibody binding site [56]. This was followed by measurements of fluorescence and photochrome parameters such as the StDNP [Pg.292]

On the basis of the available experimental and computational data, the authors came to several conclusions. The StDNP probe undergoes anisotropic rotation in the 6 cP solvent solution vith a correlation time x = l.l ns. This is much faster than theoretically predicted from the rotation barrier in vacuum about the C—N bond connecting the DNP and stilbene fragments (x,. = 1.9 ns). Therefore, the rigid label moves in solution as a vhole. [Pg.293]

These results showed that the combined fluorescence-photochrome approach can be used for investigating the local medium molecular dynamics in the immediate vicinity of specific sites of proteins and nucleic acids, as well as for other biologically important structures and synthetic analogues. [Pg.293]
