Doxepin distribution

Distribution in Blood. Plasma whole blood ratio, doxepin about 0.8 but there is considerable intersubject variation monodesmethyldoxepin about 0.6. 

The distribution of specifically bound [3H]mepyramine [99] or high-afiinity [3H]doxepin binding [105] in human brain expressed as a percentage of the binding obtained in hypothalamus. 

Its Z-N-demethylated metabolite is pharmacologically more active than its -metabolite as an inhibitor of 5-HT and NE reuptake (61). Both isomers of doxepin showed large volumes of distribution and relatively short half-lives in plasma, suggestive of extensive distribution and/or tissue binding. Renal clearances did not differ for the isomers. 

Drug overdose A fatal overdose of sulpiride and doxepin is reported in a 47-year-old female both drugs were found to be widely but differentially distributed throughout the body [278 ].
