DNA Miniprep Method

The method of Lassner et al. (1989) for minipreparations of DNA works well for transformed roots. Extraction buffer consists of 0.14M sorbitol, 0.22M Tris-HCl (pH 8), 0.022M EDTA, 0.8M NaCl, 0.8% CTAB, 1.0% 

N-laurylsarcosine and is made by mixing the CTAB (hexadecyl trimethyl ammonium bromide) and sarcosine together to form a paste before adding the rest of the solution. The EDTA is added last and the buffer is autoclaved.
