Dibenzo thiazepine

Numerous derivatives of dibenzo[/>,/][l, 4]thiazepine (1) have been tested for biological activity.

A number of monocyclic and benzo-annelated examples of 1,2- and 1,3-thiazepines have been prepared but there has been little systematic study of these systems. The interesting photochemical interconversions of pyridine N-imides into 1,2- and 1,3-diazepines and of pyridine Af-oxides into 1,2- and 1,3-oxazepines regrettably lack parallels in thiazepine chemistry. There has been more interest in 1,4-thiazepines, as both rearrangement products and possible biogenetic precursors for penicillins and because of the pharmacological value of the benzo- and dibenzo-[l,4]thiazepines as antidepressants and coronary vasodilators. The only review (70ZC361) is excellent but not very recent. 

Dibenzo[Z>,e][l,4]thiazepines have been prepared by the thermolysis of 2-azido-2, 6 -dimethyldiphenyl sulfides the reaction involves formation of the d and g bonds in a rearrangement reaction via a spirodiene intermediate (70CC233). 

Benzothiazepines of type (401) can be prepared by the Bischler-Napieralsky cycliz-ation of N- (arylthioethyl)benzamides and reactions of the same type have been used in the synthesis of dibenzo[6,/][ 1,4]thiazepines (70JHC409). 

Thioxanthen-9-one 10,10-dioxides with sodamide undergo ring expansion to dibenzo [/>,/]-[l,4]thiazepin-l 1-one 5,5-dioxides (75JHC1211). 

A review on tianeptine, a dibenzo[thiazepine derivative, has also appeared <2001MI231>. While a number of bis-fused 1,2-thiazepine systems are known, they were outside the scope of this chapter.

Dihydro-10,10-dioxo-l l-methyldibenzo[c,f][l,2]thiazepin-5-yloxy)tropane and the hydrogen maleate salt thereof. (This name was given by the authors of U. S. Patent No 3 700 633. It corresponds to endo-6,11-dihydro-6-methyl-ll-[(8-methyl-6-azabicyclo[3.2.1]oct-3-yl)oxy]dibenzo[c,f] [l,2]thiazepine 5,5-dioxide and 6,ll-dihydro-6-methyl-ll-(8-methyl-8-azabicyclo[3.2.1]octan-3a-yloxy)dibenzo[cf][l,2]thiazepin-5,5-dioxide).
