Determination of Sucrose by Invertase Hydrolase

The protein can be removed by the addition of 1N perchloric acid in the proportion of 1 1 as explained in Section 2.4.1. The solution should contain 0.4-0.8g glucose + sucrose per litre stronger solutions must be diluted. Remove excessive concentrations of glucose by treatment with glucose oxidase (Southgate 1991). [Pg.117]

Dilute 6.7 ml IM NaOH and 13.5 ml IM acetic acid with 180 ml of water. The pH should be 4.6. Bring buffer to 20-25 °C before use. (Buffer is stable for 1 year when stored at +4°C) [Pg.117]

Dissolve 10 mg of beta-fructosidase lyophilisate in 2 ml of distilled water. (Stable for 1 week when stored at +4°C) [Pg.117]

The following solution are the same as those used in Section 2.4.1. [Pg.117]

Bring buffer to 20-25 °C before use. (The buffer is stable for 4 weeks when stored at +4°C). [Pg.118]
