Certainly there are compromises that need to be made when adding complexity to an already complicated system. 

The enzyme concentration should coincide to the measurable rates of metabolism, taking into account the highest and lowest rates of metabolism seen for each enzyme. At the same time, protein content should still be kept low to minimize the compound binding [41-43]. 

An appropriate incubation time should also be evaluated to measure initial rate kinetics (depletion) of the most extensively metabolized substrate while maintaining sensitivity of the least metabolized substrate. 

Potential competition of individual isoforms for cytochrome b5 and NADPH reductase may limit the electron transport and thus metabolic rate [44, 45]. 

It has been shown that such issues can be overcome and in an acceptable range to provide an appropriate level of quality data [25, 46].
