Zinc cyclam complex

The zinc complex of the saturated macrocycle (1,2,3,4,5,6,7,8,9,10,11,12,13,14,15,16,17,18,19, 20,21,22-docosahydrodibenzo-[B,I][l,4,8,l 1] tetra-azacyclotetradecine), which is a 14[ane]N4 (cyclam) ring incorporating two cyclohexyl rings into the macrocycle, has been prepared and characterized.688 Two isomers of the ligand were separately complexed with zinc and characterized by NMR. 

Kimura and co-workers have demonstrated the potent antiviral inhibitory action of zinc complexes of bi-cyclam ligands against HIV in vitro. A series of compounds, with variation in the bridge between the macrocycles, was synthesized and tested.920... 

A number of zinc compounds and complexes have been developed for therapeutic purposes. Several zinc complexes of cyclam-based ligands have proved potent inhibitors of HIV, while hydroxypyridinone complexes are being assessed as insulin mimics for the treatment of diabetes. There is some evidence that the administration of certain zinc chelates may help to ward off strokes, while a suberoylanihde hydroxamate complex has been in clinical trials as an anticancer agent since 2000. 

Complexation and determination of formation constants has been studied with the tetramethyl-cyclam (l,4,8,ll-tetramethyl-l,4,8,ll-tetraazacyclotetradecane) ligand.686 A series of zinc N4 donor macrocyclic complexes with different ring sizes were synthesized and characterized for comparison with cadmium derivatives.687... 

The effect of several metal ions on the anti-HIV activity of bi-cyclams has been reported (371). Zinc may facilitate the binding of bicyclams to the virus, and bis-Zn(II)-bicyclam complexes exhibit activity comparable to that of the parent bicyclam, but activity is reduced for other metal complexes such as Ni(II), Cu(II), Co(II), and Pd(II).
