Cloning family 7 glycosyl hydrolase

The tomatinase gene from F. oxysporum f. sp. lycopersici has been cloned recently [34]. This gene encodes a protein that has no sequence homology to any previously described saponinase but which is highly similar to xylanases (family 10 of glycosyl hydrolases) [84, 85, 93-95]. A clone of cardenolide 16 -0-glucohydrolase cDNA (CGH I) was obtained from D. lanata. The amino acid sequence derived from CGH I showed high homology to a widely distributed family of (3-glucohydrolases (glycosyl hydrolases family 1). The recombinant CGH I protein produced in Escherichia coli had CGH 1 activity. CGH 1 mRNA was detected in leaves, flowers, stems and fruits of D. lanata (Framm et ah, 2000). [Pg.333]
