Capillary transfer large fragments

Capillary transfer has been the classic procedure but simplified modifications and different versions of vacuum blotting are becoming popular. Large fragments are not efficiently transferred and DNA is clipped into = 1 kb fragments between electrophoresis and transfer. [Pg.200]

Wahl et al. modified the gel pretreatment procedure (5), including a depurination step. The depurination breaks large fragments of DNA into smaller pieces, and this facilitates their transfer by capillary action. However, we have not found depurination to be necessary when transferring gels using the vacuum method. [Pg.26]
