Buffalo black

Amido black lOB (Bayer), or naphthalene black 12B.200 (ICI), or Pontacyl blue black SX (Du Pont), or Buffalo black (National Aniline) are most often used for routine processes. One-half gram of one of these dyes is dissolved in a solution of 5.0 g mercuric chloride and 5.0 ml acetic acid in 100.0 ml distilled water (W4). The solution is filtered before use. The staining of the slides with the agar film is done in the ordinary tray used for microscopy and takes 5 min. For differentiation, the slides are rinsed in a 2 /c solution of acetic acid in distilled water or in a water solution of 50 % methanol and 10 % acetic acid. The bath is renewed until the background is colorless. Often, new lines of precipitate show up at this stage. The slides are finally washed in water and dried within a few minutes. [Pg.227]

After electrophoretic separation, protein fractions are visualized by staining with a suitable dye. Ideally, a dye should have equal affinity for all proteins and sufficient sensitivity to detect each protein. However, the most commonly used dye for cellulose acetate separation. Ponceau S, has 1.5 times greater affinity for albumin than for y-globulin. Other widely used dyes that behave in a similar manner are Amido black 10 B (Amidoschwarz 10 B), Naphthol blue black or Buffalo black, a stain commonly employed for agarose separations, and CBB, a dye widely used for agarose and polyacrylamide gel separations. A recent study in Ethiopia found the most effective attractant for tsetse flies was acetone, octenol, and cow urine (Belete etal, 2004). An American company markets Mosquito Magnet, a device that purportedly attracts mosquitoes with l-octen-3-ol, heat, carbon dioxide, and water vapor (Enserink, 2002). Simpler traps are being used in East Africa. A black-and-blue cloth, impregnated with insecticide, attracts mosquitoes with acetone and octenol, or alternatively, buffalo urine (Enserink, 2002). [Pg.372]
