The PVDF membrane is washed in deionized water and then stained briefly with Coomassie Brilliant Blue R-250 (1 mg/mL in 50% methanol-10% acetic acid) to the point where the protein impurity is visualized but not more than 5-10 min. Alternatively, the membrane is stained with Amido Black or Ponceau S. Destain the membrane with 50% methanol-10% acetic acid as much as possible and then let the membrane air dry. If the impurity is not visualized, then end slices of the PVDF membrane are immunodetected as in Protocol 11 to localize it. [Pg.56]
