Bioavailability rigid compounds

An example compound from this series is SCFi 416538 (Fig. 3). This type of compound showed resistance to peptidases and amidases. Since a more rigid conformation is maintained, this chemotype appears to have better potency. Some compounds in this class had good oral bioavailability in rats. FFowever, the PK in dogs and monkeys was poor for most of the compounds.
