Proteins and Amino Acids Total protein in food and feed samples is commonly determined by Kjeldahl (acid digestion/titration) or Dumas (pyrolysis) or elemental analysis.14 FIPLC can separate major proteins and furnish protein profiles and speciation information. HPLC can be used to further characterize specific proteins via peptide mapping and amino acid sequence analysis. HPLC modes used for protein include IEC, SEC, RPC, and affinity chromatography with typical UV detection at 215 nm or MS analysis. Some amino acids are more abundant in proteins than other amino acids. Cysteine, tryptophan, and methionine are rare amino acids together they constitute approximately 5 percent of the amino acids in a protein. Four amino acids— leucine, serine, lysine, and glutamic acid—are the most abundant amino acids, totaling 32 percent of all the amino acid residues in a typical protein. However, the amino acid composition of proteins can vary widely from these values. 

The extraction of iodine amino acids from the blood serum (butanol-extractable iodine, BEI) showed that approximately 85% of the FBI consisted of iodine amino acids. Total iodine, FBI and BEI in the serum of farm animals was correlated (r = 0.83 and r = 0.89, respectively) (Groppel 1986). However, no correlation was observed between the total serum iodine content and T4 correlated with r = 0.66.
