Abrin chromatography

Abrin A and Abrin B. Toxic proteins from seeds of Abras precatorius. Purified by successive chromatography on DEAE-Sephadex A-50, carboxymethylcellulose, and DEAE-cellulose. [Wei et al. JBC 249 3061 J974].

The reaction mixture obtained following the conjugation of a mouse monoclonal antibody (50 mg) and [l25I]-labeled abrin A chain was chromatographed on a column of Sephacryl S-200 (SF), dimensions 80 cm x 2.6 cm (id). Fractions eluting from the column were monitored spectrophotometrically at 280 nm (—) to measure total protein, and by gamma counting (—) to measure the A chain in its free or conjugated form. The hatched area indicates a typical pooled conjugate preparation.

For the purification of antibody raised against ridn and abrin A chains, a useful alternative to affinity chromatography on columns made with the isolated A chains is the use of immobilized /h dnus and Abrus agglutinins, which crossreact immunologically with the corresponding toxins but are much less toxic.
