# Cardinality

Sometimes the results of QRA will be used in the public arena, and communicating to the public about the risks of exposure to chemicals is difficult. You must be sensitive to the feelings of a public that is generally suspicious of industry and ignorant of science. Streamlining has a major effect on the energy efficiency of bodies moving through a fluid. For example, a bicycle with its odd shaped surfaces, has a relatively large drag coefficient. In contrast, a streamlined outer shell used for recumbent bicycles reduces the drag and has allowed the speed record to reach 67 mph. Streamlining is a cardinal principle in airplane design, where drag reduction is so important. [c.13]

Cleavage of the 8-lactam carbonyl-nitrogen bond is of cardinal significance, because the antibacterial activity of 8-lactam antibiotics is dependent on this reaction. The ability of 8-lactam antibiotics to act as acylating agents within the active site of bacterial enzymes critical for cell wall biosynthesis is, in turn, dependent on the reactivity of the 8-lactam carbonyl. At the same time the carbonyl must not be so reactive that it is hydrolyzed by adventitious nucleophiles prior to its arrival at the target enzymes. Thus, the proper balance between reactivity and stability of the 8-lactam ring must be struck to achieve maximum antibacterial potency. Furthermore, since the more reactive 8-lactams are cleaved under relatively mild conditions, this feature must be carefully considered in any strategy for the total synthesis or chemical modification of penicillins and cephalosporins. Compared to penicillins, cephalosporins are relatively more stable to 8-lactam cleavage. Thus, while penicillins are normally stable only near neutrality (pH 5-8) and react readily with alcohols, cephalosporins are hydrolyzed less readily, particularly under acid conditions, and have sufficient stability to permit their recrystallization from methanol. However, the P-lactam of cephalosporin does react readily with good nucleophiles. For example, cleavage by hydroxylamine to form a hydroxamic acid is the basis of a rapid assay for p-lactam antibiotics. Reaction is essentially complete within 3 min at pH 7 (B-72MI51003). [c.287]
